Beyond redox chemistry, NAD+ is consumed as a substrate by enzymes that transfer ADP-ribose or remove acetyl groups. Sirtuins use NAD+ in deacylation reactions, poly(ADP-ribose) polymerases use it in DNA damage responses, and CD38 hydrolases convert it to signaling metabolites. Because these enzymes compete for the same pool, changes in NAD+ availability can influence multiple cellular processes. The relative contribution of each consumption route differs by cell type and condition, and precise quantitative links remain an active area of study.
Research on NAD+ spans biochemistry, aging biology, and metabolism. Studies often examine how NAD+ levels change with age, diet, exercise, or disease states, and whether precursor supplementation alters those levels. Findings in animal models do not automatically translate to humans, and measurement methods vary across studies. Questions about tissue-specific effects, long-term consequences, and causal relationships remain open. NAD+ itself is not established as a single therapeutic agent with a broad clinical role.
Laboratory handling of NAD+ follows standard practices for hygroscopic fine chemicals. Personnel typically avoid inhalation and skin contact, use gloves and eye protection, and work in a ventilated area. Quality control may include ultraviolet absorbance at the nicotinamide maximum, chromatographic purity, water content, and identity confirmation by mass spectrometry. Because commercial preparations can contain counterions, residual solvents, or related nucleotides, a certificate of analysis helps verify the material. Researchers should confirm that the form supplied matches the intended assay.
Measuring NAD+ in biological samples requires care because the molecule is chemically reactive and present at low concentrations in some tissues. Common approaches include enzymatic cycling assays, high-performance liquid chromatography, and liquid chromatography coupled to mass spectrometry. Each method has different sensitivity and specificity, and sample preparation can affect results. Acidic or alkaline extraction steps are used in some protocols, but the choice depends on the analyte and matrix. No single method is universally optimal for every tissue or fluid.

Cells produce NAD+ through several biosynthetic routes. The salvage pathway recycles nicotinamide, while the Preiss-Handler pathway uses nicotinic acid, and a de novo route can start from tryptophan in some organisms. In mammals, the salvage pathway is generally considered the main source under ordinary conditions. Tissue concentrations vary widely by cell type and compartment, and measured declines with age have been reported in some studies. Whether such changes drive aging or mainly accompany it remains an open question.
Nicotinamide adenine dinucleotide, commonly abbreviated NAD+, is a coenzyme present in all living cells. The molecule consists of two nucleotides linked by phosphate groups, with adenine and a nicotinamide ring as its principal features. In its oxidized form, the nicotinamide ring can accept a hydride ion, becoming NADH. This reversible conversion places NAD+ at the center of many electron-transfer reactions. Its role as a redox carrier is well established across bacteria, plants, fungi, and animals.
Commercial NAD+ is supplied as a solid, often as the free acid or a salt, and purity is verified by chromatographic methods. Laboratories typically store it desiccated at minus 20 degrees Celsius or below. Working solutions are prepared fresh because even sterile aqueous solutions can lose activity over hours to days depending on pH and temperature. Documentation may include a certificate of analysis, an assay value, and a recommended retest date. Researchers should verify identity and purity when results depend on precise cofactor concentrations.
NAD+ is commonly measured by high-performance liquid chromatography with ultraviolet detection, often at 254 or 260 nm. Enzymatic cycling assays provide higher sensitivity by coupling NAD+ to a reporter reaction. Mass spectrometry can distinguish NAD+ from close analogues and confirm isotope labeling. Sample preparation usually involves rapid quenching of metabolism to prevent interconversion with NADH. Because NAD+ and NADH differ by one hydride, extraction conditions strongly affect the measured ratio.
In aqueous solution, NAD+ is most stable under mildly acidic to neutral conditions and degrades faster at high pH or elevated temperature. The molecule can hydrolyze at the pyrophosphate bond or undergo nonenzymatic cyclization. Buffers, chelating agents, and cold temperatures slow these losses during analysis. Repeated freeze-thaw cycles are generally avoided because they can promote degradation and concentration changes. Light exposure is also controlled, though NAD+ is less photolabile than some related nucleotides.
NAD+ also serves as a substrate for enzymes that cleave it, including sirtuins, PARPs, and CD38. These enzymes consume NAD+ and release nicotinamide and ADP-ribose or related products. The dual roles as redox cofactor and signaling substrate connect NAD+ to DNA repair, circadian regulation, and calcium signaling. Cellular NAD+ concentrations vary by tissue, time of day, and stress exposure. How these consumption pathways interact with redox balance remains an active area of research.
NAD+ is a dinucleotide composed of two nucleotides joined by a pyrophosphate linkage. One nucleotide contains adenine, and the other contains nicotinamide. The oxidized form carries a positive charge on the nicotinamide ring and is abbreviated NAD+. It functions as a cofactor in hydride-transfer reactions, accepting electrons in catabolic pathways. In cells, it interconverts with reduced NADH, forming a redox couple central to energy metabolism. The molecule is water-soluble and does not cross cell membranes freely without specific transport or precursor pathways.
The nicotinamide ring undergoes reversible reduction at the para position, converting NAD+ to NADH. This reaction transfers a hydride equivalent, not a free hydrogen atom or electron alone. Because the redox pair has a defined reduction potential, it links oxidation of fuels to respiratory chain activity. Many dehydrogenases use NAD+ as a co-substrate and produce NADH. The ratio of NAD+ to NADH reflects metabolic state and influences flux through several pathways.
=== In U.S. drinking water === In 2000, perchlorate contamination beneath the former flare manufacturing plant Olin Corporation Flare Facility, Morgan Hill, California was first discovered several years after the plant had closed. The plant had used potassium perchlorate as one of the ingredients during its 40 years of operation. By late 2003, the State of California and the Santa Clara Valley Water District had confirmed a groundwater plume currently extending over nine miles through residential and agricultural communities. The California Regional Water Quality Control Board and the Santa Clara Valley Water District have engaged in a major outreach effort, a water well testing program has been underway for about 1,200 residential, municipal, and agricultural wells. Large ion exchange treatment units are operating in three public water supply systems which include seven municipal wells with perchlorate detection. The potentially responsible parties, Olin Corporation and Standard Fuse Incorporated, have been supplying bottled water to nearly 800 households with private wells, and the Regional Water Quality Control Board has been overseeing cleanup efforts. The source of perchlorate in California was mainly attributed to two manufacturers in the southeast portion of the Las Vegas Valley in Nevada, where perchlorate has been produced for industrial use.

== Normal values == Note: mm/h. = millimeters per hour. Westergren's original normal values (men 3 mm/h and women 7 mm/h) made no allowance for a person's age. Later studies from 1967 confirmed that ESR values tend to rise with age and to be generally higher in women. Values of the ESR also appear to be slightly higher in normal populations of African-Americans than Caucasians of both genders. Values also appear to be higher in anemic individuals than non-anemic individuals.

This work hinged on his demonstration that red blood cell glutathione was unstable to oxidative stress. Later, he was to develop an assay for glutathione that was widely used in studies of red cell oxidative metabolism. He was later transferred to Camp Detrick in Frederick, Maryland (1954–1955), where he studied Q fever. He was honorably discharged from the Army with the rank of captain. Beutler then joined the faculty of the Department of Medicine at the University of Chicago, where he studied iron metabolism and red blood cell metabolism. In 1959, he became chairman of the Department of Medicine of the City of Hope National Medical Center in Duarte, California, and in 1979 assumed the chairmanship of the Department of Clinical Research at the Scripps Clinic and Research Foundation. Three years later, he was asked to become Chairman of a merged department (the Department of Molecular and Experimental Medicine) at Scripps, which later became The Scripps Research Institute in La Jolla, CA. He maintained his position as Chairman until his 80th birthday, only a few days before his death. Not long after moving to California, Beutler made one of his most important contributions. A new colleague at the City of Hope and ultimately a lifelong friend, Susumu Ohno had recently demonstrated that the histologically observable Barr body present in the nuclei of mammalian female cells was a hyperchromatic X chromosome. Beutler immediately recognized that this might account for the variable expression of X-linked genes in females heterozygous for X-linked mutations.
However, confirmation of the acyl enzyme was done without trapping experiments, making the conclusions weak. The second proposed mechanism is a promoted water pathway. This mechanism involves attack of a water molecule at the scissile peptide linkage of the substrate. This process is promoted by the zinc ion and assisted by residue Glu-270.
When analyte molecules with electronegative / withdrawing elements or functional groups electrons are captured which results in a decrease in current generating a detector response. Nitrogen–phosphorus detector (NPD), a form of thermionic detector where nitrogen and phosphorus alter the work function on a specially coated bead and a resulting current is measured. Dry electrolytic conductivity detector (DELCD) uses an air phase and high temperature (v. Coulsen) to measure chlorinated compounds. Mass spectrometer (MS), also called GC-MS; highly effective and sensitive, even in a small quantity of sample. This detector can be used to identify the analytes in chromatograms by their mass spectrum. Some GC-MS are connected to an NMR spectrometer which acts as a backup detector. This combination is known as GC-MS-NMR. Some GC-MS-NMR are connected to an infrared spectrophotometer which acts as a backup detector. This combination is known as GC-MS-NMR-IR. It must, however, be stressed this is very rare as most analyses needed can be concluded via purely GC-MS. Vacuum ultraviolet (VUV) represents the most recent development in gas chromatography detectors. Most chemical species absorb and have unique gas phase absorption cross sections in the approximately 120–240 nm VUV wavelength range monitored. Where absorption cross sections are known for analytes, the VUV detector is capable of absolute determination (without calibration) of the number of molecules present in the flow cell in the absence of chemical interferences.
== Serving and consumption == In order to properly honor the transition from preparation to consumption, the Inca used ceramic vessels as a way to add artistic and cultural value to the dishes being served. Present at archaeological excavations of Machu Picchu are the ceramic vessels known as cazuelas, prominent among others found at the site. Archaeologists interpreted that the vessel was used for serving, rather than preparation, due to its morphology. With its openness, the vessel is easily accessible for consumption purposes, and lacks aspects that support pouring or closing. Cazuelas could have been used for consumption of soups, stews, porridges, or even chicha. Its characteristic polychrome designs further support idea of the vessel being used for serving and consumption purposes. With the cultural significance held by chicha, it had an array of purposes when it came to consumption. Being used in ceremonial and ritual events, chicha was used to influence social relations and classes. Chicha was used as a way to mitigate relationships between authoritative figures and workers, especially in the workplace itself. Toasting of chichawas often associated with connection to honoring ancestors. Consumption could either be done at large-scale gatherings, or in private settings, which were more restricted to elite members of the community. Specifically, through use of isotope analysis, archaeologists have discovered a relationship between maize consumption and status of combatants who lived in Majes Valley, a pre-hispanic region located in the Andes.

=== DES litigation === Lilly was one of numerous manufacturers of diethylstilbestrol (DES), a synthetic estrogen that was prescribed to pregnant women to prevent pregnancy complications such as miscarriage. In 1971, researchers linked prenatal DES exposure to clear-cell adenocarcinoma of the vagina and cervix. The Food and Drug Administration responded by advising physicians not to prescribe DES to pregnant women. More than 300 companies manufactured DES, which later complicated litigation because plaintiffs often could not identify which manufacturer had supplied the drug taken by their mothers. In Bichler v. Eli Lilly and Company, Joyce Bichler sued Lilly after developing cervical and vaginal cancer at age 17 following prenatal exposure to DES. In 1982, the New York Court of Appeals upheld a $500,000 jury award to Bichler even though it had not been established that Lilly manufactured the particular DES taken by her mother. In the 1989 case Hymowitz v. Eli Lilly and Company, the New York Court of Appeals adopted a form of market share liability for DES cases that allowed liability to be apportioned among manufacturers according to their share of the national DES market when the specific producer could not be identified. DES litigation continued for decades. In 2013, Lilly reached an undisclosed settlement during a federal trial brought by four sisters who alleged that their breast cancers resulted from DES their mother had taken during pregnancy. Lilly disputed that DES caused their cancers and disputed whether their mother had taken DES manufactured by Lilly.
=== East vs West === The dissonance between Japanese tradition and Western modernity is present throughout the novel. Kaname's aesthetic tastes lean more toward the West. His romanticized version of it is manifested in the western wing of his house, in particular the veranda on which he likes to sit, in his fascination with American movie stars, and in his skimming through an English translation of the Arabian Nights for lewd passages. However, after a visit to the bunraku theater with his wife, his father-in-law, and his father-in-law's mistress in Chapter Two, Kaname's interest in traditional aesthetics is piqued, and he even becomes envious of the "old man" and his lifestyle: watching an old play, pipe in hand, sake and a young mistress at his side. This is the beginning of Kaname's divergent interest in the East, his preference for the past.
== Food chain versus food web == A food chain differs from a food web as a food chain follows a direct linear pathway of consumption and energy transfer. Natural interconnections between food chains make a food web, which are non-linear and depict interconnecting pathways of consumption and energy transfer.
In both males and females, FSH stimulates the maturation of primordial germ cells. In males, FSH induces Sertoli cells to secrete androgen-binding proteins (ABPs), regulated by inhibin's negative feedback mechanism on the anterior pituitary. Specifically, activation of Sertoli cells by FSH sustains spermatogenesis and stimulates inhibin B secretion. In females, FSH initiates follicular growth, specifically affecting granulosa cells. With the concomitant rise in inhibin B, FSH levels then decline in the late follicular phase. This seems to be critical in selecting only the most advanced follicle to proceed to ovulation. At the end of the luteal phase, there is a slight rise in FSH that seems to be of importance to start the next ovulatory cycle. Control of FSH release from the pituitary gland is unknown. Low frequency gonadotropin-releasing hormone (GnRH) pulses increase FSH mRNA levels in the rat, but is not directly correlated with an increase in circulating FSH. GnRH has been shown to play an important role in the secretion of FSH, with hypothalamic–pituitary disconnection leading to a cessation of FSH. GnRH administration leads to a return of FSH secretion. FSH is subject to oestrogen feed-back from the gonads via the hypothalamic pituitary gonadal axis.
As of 2024, it is unclear whether (S)-tetrahydropapaverine derives from (S)-reticuline or (S)-coclaurine: the difference is whether the piperidine nitrogen atom is methylated at any point during the biosynthetic sequence.
