Beyond redox chemistry, NAD+ is consumed as a substrate by enzymes that transfer ADP-ribose or remove acetyl groups. Sirtuins use NAD+ in deacylation reactions, poly(ADP-ribose) polymerases use it in DNA damage responses, and CD38 hydrolases convert it to signaling metabolites. Because these enzymes compete for the same pool, changes in NAD+ availability can influence multiple cellular processes. The relative contribution of each consumption route differs by cell type and condition, and precise quantitative links remain an active area of study.
Research on NAD+ spans biochemistry, aging biology, and metabolism. Studies often examine how NAD+ levels change with age, diet, exercise, or disease states, and whether precursor supplementation alters those levels. Findings in animal models do not automatically translate to humans, and measurement methods vary across studies. Questions about tissue-specific effects, long-term consequences, and causal relationships remain open. NAD+ itself is not established as a single therapeutic agent with a broad clinical role.
Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide composed of two nucleotides joined by phosphate groups. One nucleotide contains adenine; the other contains nicotinamide. The molecule exists in oxidized (NAD+) and reduced (NADH) forms, and the reversible hydride transfer between them underlies many metabolic oxidation-reduction reactions. In cells, NAD+ serves as an electron acceptor in pathways such as glycolysis, the citric acid cycle, and oxidative phosphorylation. Its concentration and redox ratio vary by compartment, tissue, and metabolic state.
Stability studies show that NAD+ can hydrolyze under prolonged heat, extreme pH, or microbial contamination. Phosphate buffers near neutral pH are often used for short-term handling, though exact stability depends on concentration, temperature, and matrix. In biological samples, endogenous enzymes can rapidly degrade NAD+, making cold chain and fast processing important. Analytical reports should state extraction conditions, internal standards, and validation parameters. Without those details, comparisons across studies remain difficult and potentially misleading.
Laboratory measurement of NAD+ often begins with rapid quenching of cell or tissue samples to prevent enzymatic conversion. Acidic or alkaline extraction can precipitate proteins, but the chosen method affects recovery of oxidized and reduced forms. Enzymatic cycling assays provide high sensitivity by amplifying a NAD+-dependent reaction. High-performance liquid chromatography and mass spectrometry offer separation and structural confirmation. Each method has trade-offs in throughput, specificity, and the ability to distinguish NAD+ from close analogues.

Quality control for NAD+ relies on identity, purity, and functional tests. A certificate of analysis may report high-performance liquid chromatography purity, ultraviolet spectrum, water content, and residual solvents. Because NAD+ is hygroscopic, gravimetric values can shift as material absorbs water, so purity should be interpreted alongside storage history. Mass spectrometry confirms molecular identity, while enzymatic assays show whether the material supports dehydrogenase activity. Commercial material is available as the free acid and as salts, and the counterion affects molecular weight, solubility, and how concentrations are calculated.
Laboratory measurement of NAD+ usually begins with rapid sample quenching because the molecule can change form after collection. Enzymatic cycling assays amplify signal through coupled reactions and are suited to small samples. High-performance liquid chromatography with ultraviolet detection separates NAD+ from related nucleotides. Liquid chromatography-mass spectrometry offers higher specificity and can distinguish NAD+ from close analogs. Each method has trade-offs in sensitivity, throughput, and equipment needs, so reported values depend heavily on extraction and detection choices.
The stability of NAD+ depends on pH, temperature, light exposure, and the presence of degradative enzymes. Aqueous solutions are generally more stable under mildly acidic to neutral conditions and degrade faster under alkaline conditions or prolonged heat. The solid is hygroscopic and should be stored desiccated, often frozen, and protected from repeated freeze-thaw cycles. In laboratory handling, aliquots reduce repeated temperature changes, and chelating agents may limit metal-catalyzed hydrolysis in some buffers. These practices matter because even small amounts of NADH or hydrolysis products can interfere with quantitative assays.
Quality control for NAD+ materials typically combines identity, purity, and water content checks. Identity may be confirmed by ultraviolet spectrum, retention time in chromatography, or mass accuracy, while purity is assessed by HPLC peak area or quantitative nuclear magnetic resonance. Residual water and solvents can affect molar calculations and enzyme assays, so Karl Fischer titration or thermogravimetric analysis may be used. Commercial materials vary in grade and counterion form, and published methods should specify the exact salt or hydrate when reporting concentrations. Regulatory status depends on intended use, with research reagents, dietary ingredients, and clinical products treated under different frameworks.
Quantification of NAD+ in biological samples usually relies on separation techniques coupled to sensitive detection. High-performance liquid chromatography with ultraviolet detection can measure the oxidized form by its absorbance near 260 nm, while mass spectrometry provides greater specificity and can distinguish NAD+ from close analogs. Enzymatic cycling assays use coupled dehydrogenase reactions to amplify signal and estimate NAD+ concentrations in cell or tissue extracts. Because NAD+ and NADH interconvert rapidly, sample preparation must quench metabolism quickly and preserve the redox state before analysis.
Measuring NAD+ in biological samples requires care because the molecule is chemically reactive and present at low concentrations in some tissues. Common approaches include enzymatic cycling assays, high-performance liquid chromatography, and liquid chromatography coupled to mass spectrometry. Each method has different sensitivity and specificity, and sample preparation can affect results. Acidic or alkaline extraction steps are used in some protocols, but the choice depends on the analyte and matrix. No single method is universally optimal for every tissue or fluid.
Solid NAD+ is relatively stable when kept dry, cold, and protected from light. Aqueous solutions are more vulnerable to hydrolysis and can lose activity during repeated freeze-thaw cycles or prolonged storage at ambient temperature. Stability depends on pH, ionic strength, and the presence of degrading enzymes or metal ions. For many laboratory uses, aliquots are stored frozen and thawed only once. Exact degradation rates vary by matrix, so stability should be checked for each application rather than assumed.
Beyond redox catalysis, NAD+ is a substrate for enzymes that transfer ADP-ribose or remove acetyl groups from proteins. Sirtuins and poly(ADP-ribose) polymerases consume NAD+ and release nicotinamide as a byproduct. These reactions connect cellular energy status to gene regulation, DNA repair, and stress responses. Because NAD+ is used rather than merely recycled in such signaling, its concentration reflects both biosynthesis and consumption. The balance between salvage and de novo synthesis pathways determines available pools in different tissues.
Biosynthesis of NAD+ starts from nicotinamide, nicotinic acid, or nicotinamide riboside through salvage pathways. A rate-limiting enzyme, nicotinamide phosphoribosyltransferase, converts nicotinamide to nicotinamide mononucleotide. Further coupling with ATP yields NAD+. In mammals, the liver and muscle can synthesize NAD+ from dietary precursors, but tissue levels vary widely. Researchers study these pathways to understand age-related changes, metabolic disorders, and neurodegeneration. Direct causal links between NAD+ decline and disease remain an active area of investigation.
NAD+ is a dinucleotide composed of adenine, ribose, and nicotinamide groups joined by phosphate linkages. It serves as a coenzyme in oxidoreductase reactions, cycling between oxidized NAD+ and reduced NADH. The molecule is water-soluble and occurs in all living cells. Its nicotinamide ring accepts hydride ions during catabolic reactions, linking substrate oxidation to electron transport. This redox couple supports ATP production and helps maintain cytosolic and mitochondrial redox balance in many cell types.
== Histology == The epididymis is covered by a two-layered pseudostratified epithelium. The epithelium is separated by a basement membrane from the connective tissue wall, which has smooth muscle cells. The major cell types in the epithelium are:
The formation of these linear chains appears to be driven by the development of an electrostatic dipole in each of the colloidal spheres strong enough to overcome coulomb repulsion. This suggests a possible mechanism by which alpha sheet may promote amyloid aggregation; the peptide bond has a relatively large intrinsic electrostatic dipole, but normally the dipoles of nearby bonds cancel each other out. In the alpha sheet, unlike other conformations, the peptide bonds are oriented in parallel so that the dipoles of the individual bonds can add up to create a strong overall electrostatic dipole. Notably, the protein lysozyme is among the few native-state proteins shown to contain an alpha-strand region; lysozyme from both chickens and humans contains an alpha strand located close to the site of a mutation known to cause hereditary amyloidosis in humans, usually an autosomal dominant genetic disease. Molecular dynamics simulations of the mutant protein reveal that the region around the mutation assumes an alpha strand conformation. Lysozyme is among the naturally occurring proteins known to form amyloid fibers under experimental conditions, and both natively alpha-strand region and the mutation site fall within the larger region identified as the core of lysozyme amyloid fibrillogenesis. A mechanism for direct alpha sheet and beta sheet interconversion has also been suggested, based on peptide plane flipping in which the αRαL dipeptide inverts to produce a ββ dihedral angle conformation.
While American slaves in the Antebellum South were sold for around $40,000 (in inflation adjusted dollars), a slave nowadays can be bought for just $90, making replacement more economical than providing long-term care. Slavery is a multibillion-dollar industry with estimates of up to $35 billion generated annually.

== The Establishment of Institute of Biological Chemistry, Academia Sinica == Despite his significant scientific achievements, Dr. Li was also dedicated to the initiation and development of protein research in Taiwan. In spring 1958, Dr. Li was invited by Dr. Shih Hu to lecture a three-week course regarding the newly developed technology in protein chemistry and his latest pituitary gland research at National Taiwan University with the support from China Foundation for the Promotion of Education and Culture. At that time, the academic community only knew that protein research was developing rapidly abroad and a scholar had already made extraordinary achievements, and so when Dr. Li introduced the first-hand knowledge, it really attracted and inspired domestic scientists. Dr. Li believed that the protein chemistry was the foundation of future biochemical and biological sciences research, so he decided to assist the establishment of domestic protein research institute. In order to cultivate talents in Taiwan, Dr. Li personally provided scholarships and selected and persuaded scholars who have settled in Taiwan to continue the research in his or other research institutes. With the support from Academia Sinica, National Science Council, Ministry of Education, and National Taiwan University, Institute of Biological Chemistry and Institute of Biochemical Sciences, College of Life Science, National Taiwan University were established in 1972.

== Litigation == CLIA itself has not been the subject of much litigation. Federal district courts have held that the CLIA did not create a private cause of action for individuals to sue laboratories that do not comply with its provisions.
Symptoms of overdose include respiratory depression, drowsiness leading to coma and sometimes to death, drooping of skeletal muscles, low heart rate, and decreasing blood pressure. At the hospital, individuals with hydromorphone overdose are provided supportive care, such as assisted ventilation to provide oxygen and gut decontamination using activated charcoal through a nasogastric tube. Opioid antagonists, such as naloxone, also may be administered concurrently with oxygen supplementation. Naloxone works by reversing the effects of hydromorphone, and is administered only in the presence of significant respiratory depression and circulatory depression. Sugar cravings associated with hydromorphone use are the result of a glucose crash after transient hyperglycemia following injection, or a less profound lowering of blood sugar over a period of hours, in common with morphine, heroin, codeine, and other opioids.

=== Rheumatoid arthritis === Adalimumab has been shown to reduce the signs and symptoms of moderate to severe rheumatoid arthritis in adults. It may be used alone or in combination with disease-modifying antirheumatic drugs. It has also been shown to have efficacy in moderate to severe polyarticular juvenile idiopathic arthritis in children four years and older, and is indicated for the treatment of that condition. In rheumatoid arthritis, it is indicated for use alone, or with methotrexate or similar medicines, in the United States since 2002. It has a similar effectiveness as methotrexate and, in combination, nearly doubles the response rate of methotrexate alone.
We will keep pressing. We will keep pushing, keep advancing, no quarter, no mercy for our enemies. Yet some in this crew, in the press, just can't stop. Allow me to make a few suggestions. People look up at the TV and they see banners, they see headlines. I used to be in that business. And I know that everything is written intentionally. For example, a banner or a headline: "Mideast war intensifies," splashing on the screen the last couple of days, alongside visuals of civilian or energy targets that Iran has hit, because that's what they do. What should the banner read instead? How about, 'Iran increasingly desperate,' because they are. They know it and so do you, if it can be admitted. Or more fake news from CNN, "reports that the Trump administration underestimated the Iran war's impact on the Strait of Hormuz" – patently ridiculous, of course. For decades, Iran has threatened shipping in the Strait of Hormuz. This is always what they do, hold the strait hostage. CNN doesn't think we thought of that. It's a fundamentally unserious report. The sooner David Ellison takes over that network, the better.Another example of a fake headline that I saw yesterday, "war widening." Here's a real headline for you, for an actual patriotic press: how about, 'Iran shrinking, going underground'? You see, Iran's leaders are hiding in bunkers and moving into civilian areas. The only thing that is widening is our advantage, not to mention our Gulf partners stepping up even more, now going on the offense, and have always been with us on the defense with collective and integrated air defenses.
=== Circulatory system === Earthworms have a dual circulatory system in which both the coelomic fluid and a closed circulatory system carry the food, waste, and respiratory gases. The closed circulatory system has five main blood vessels: the dorsal (top) vessel, which runs above the digestive tract; the ventral (bottom) vessel, which runs below the digestive tract; the subneural vessel, which runs below the ventral nerve cord; and two lateroneural vessels on either side of the nerve cord. The dorsal vessel is mainly a collecting structure in the intestinal region. It receives a pair of commissural and dorsal intestines in each segment. The ventral vessel branches off to a pair of ventro-tegumentaries and ventro-intestinals in each segment. The subneural vessel also gives out a pair of commissurals running along the posterior surface of the septum. The pumping action on the dorsal vessel moves the blood forward, while the other four longitudinal vessels carry the blood rearward. In segments seven through eleven, a pair of aortic arches ring the coelom and acts as hearts, pumping the blood to the ventral vessel that acts as the aorta. The blood consists of ameboid cells (hemocytes) and haemoglobin dissolved in the plasma. The second circulatory system derives from the chloragogen cells of the digestive system that line the coelom. As the digestive cells become full, they release chloragocytes into the fluid-filled coelom, where they float freely but can pass through the walls separating each segment, moving food to other parts and assist in immunity and wound healing.
Protective clothing is designed to protect operators in the event of a moving chain touching their clothing by snarling the chain and sprocket, by using special synthetic fibers woven into the garment. Despite safety features and protective clothing, injuries can still occur, due to the large forces involved in the work from the fast-moving, sharp chain, or the vibration and noise of the machinery. A common accident arises from "kickback" when a chain tooth at the tip of the guide bar catches on wood without cutting through it. This throws the bar (with its moving chain) in an upward arc toward the operator, which can cause serious injury or even death. A dangerous situation occurs when heavy timber begins to fall or move before a cut is complete. The chainsaw operator may be trapped or crushed. Similarly, timber falling in an unplanned direction may harm the operator or other workers, or an operator working at a height may fall or be injured by falling timber. Like other hand-held machinery, the operation of chainsaws can cause vibration white finger, tinnitus, or industrial deafness. These symptoms were very common before vibration damping using rubber or steel springs were introduced. Heated handles are additional help. Newer, lighter, and easier to wield cordless electric chainsaws use brushless motors, which further decrease noise and vibration compared to traditional petroleum-powered models. The risks associated with chainsaw use mean that protective clothing like chainsaw boots, chaps, and hearing protectors are normally worn while cutting.

NAD+ is a coenzyme found in all living cells. It carries electrons in metabolic reactions and also serves as a substrate for enzymes involved in signaling and DNA repair. Its oxidized and reduced forms are central to energy metabolism.
NAD+ is the oxidized form and NADH is the reduced form. The pair accepts and donates electrons in redox reactions. Their ratio helps indicate the metabolic state of a cell or compartment.
No. Nicotinamide mononucleotide (NMN) and nicotinamide riboside (NR) are precursors that cells can convert into NAD+. They are distinct molecules with different absorption and metabolism profiles.
Many enzymes consume or produce NAD+ within seconds after a sample is collected. Quenching stops those reactions and helps preserve the ratio between oxidized and reduced forms. The exact quenching method depends on the tissue or cell type and the analytes of interest.
