Beyond redox chemistry, NAD+ serves as a substrate for enzymes that cleave the molecule and transfer its ADP-ribose moiety or remove acetyl groups. Sirtuins consume NAD+ during deacetylation, poly(ADP-ribose) polymerases use it in DNA damage responses, and CD38 enzymes hydrolyze it to signaling metabolites. These consumption pathways mean that NAD+ availability can influence gene regulation, DNA repair, and calcium signaling. Cellular NAD+ concentrations decline in some tissues with age in animal models, but whether this decline is a cause or consequence of aging in humans remains an active open question.
Nicotinamide adenine dinucleotide, commonly abbreviated NAD+, is a dinucleotide coenzyme built from an adenine nucleotide and a nicotinamide nucleotide joined by a pyrophosphate linkage. Its oxidized form carries a positive charge on the nicotinamide ring, while the reduced form, NADH, carries a hydride equivalent. The molecule participates in hundreds of oxidoreductase reactions, where it accepts or donates electrons and protons. Because it can cycle between oxidized and reduced states without net consumption, NAD+ functions as a reusable electron carrier rather than a fuel molecule.
Commercial NAD+ is available at research grade, often with purity specifications determined by high-performance liquid chromatography. Certificates of analysis may report water content, residual solvents, and counterion identity. Identity can be confirmed by ultraviolet absorbance near 260 nm, mass spectrometry, or enzymatic activity. Because different salt forms and hydration states exist, researchers should verify that the product matches the intended molecular form. Lot-to-lot variation in purity can affect quantitative assays and should be documented.
Solid NAD+ is usually supplied as a white to off-white powder or lyophilized preparation. It is hygroscopic and should be kept desiccated at low temperature, commonly -20 °C or below for long-term storage. Aqueous solutions are less stable than dry material and are often prepared fresh or stored frozen in aliquots. Light exposure and repeated freeze-thaw cycles can promote degradation, so amber containers and single-use aliquots are preferred. Buffered solutions near neutral pH are generally less stable than acidic or frozen preparations.

Beyond electron transfer, NAD+ serves as a substrate for enzymes that cleave the molecule and transfer ADP-ribose units. Sirtuins, poly(ADP-ribose) polymerases, and CD38 consume NAD+ in regulatory reactions. These activities link NAD+ availability to DNA repair, chromatin modification, calcium signaling, and metabolic stress responses. Because consumption can exceed biosynthesis under some conditions, cellular NAD+ levels are dynamic rather than fixed. Enzyme affinity and local synthesis also influence how much NAD+ is available for signaling.
NAD+ is a dinucleotide composed of adenine, ribose, and nicotinamide moieties linked by phosphate groups. Its oxidized form carries a positive charge on the nicotinamide ring, which enables reversible hydride transfer. The molecule functions as a coenzyme in oxidoreductase reactions rather than as a dietary vitamin in its intact form. Cells maintain separate pools in cytoplasm, mitochondria, and nucleus. This compartmentalization allows distinct redox environments while preserving a shared chemical identity.
In glycolysis, NAD+ accepts electrons during the oxidation of glyceraldehyde-3-phosphate, forming NADH. The tricarboxylic acid cycle and fatty acid oxidation also generate NADH, which donates electrons to the mitochondrial electron transport chain. This flow supports ATP synthesis and helps maintain the redox balance of the cell. Other dehydrogenases use NAD+ as a cofactor for biosynthetic reductions and detoxification reactions. NADH is later reoxidized to sustain continued flux through these pathways.
Stability of NAD+ depends on form, temperature, pH, and water content. The solid is generally more stable than solutions, and it should be kept dry and cold. In solution, hydrolysis can cleave the dinucleotide, especially under alkaline conditions or at elevated temperature. Light exposure may also contribute to degradation. Buffers, chelating agents, and sterile handling can reduce losses, but no single condition preserves all preparations indefinitely. Researchers often prepare working solutions shortly before use and verify activity or purity after storage.
Quality control for NAD+ relies on identity, purity, and functional tests. A certificate of analysis may report high-performance liquid chromatography purity, ultraviolet spectrum, water content, and residual solvents. Because NAD+ is hygroscopic, gravimetric values can shift as material absorbs water, so purity should be interpreted alongside storage history. Mass spectrometry confirms molecular identity, while enzymatic assays show whether the material supports dehydrogenase activity. Commercial material is available as the free acid and as salts, and the counterion affects molecular weight, solubility, and how concentrations are calculated.
Laboratory measurement of NAD+ usually begins with rapid sample quenching because the molecule can change form after collection. Enzymatic cycling assays amplify signal through coupled reactions and are suited to small samples. High-performance liquid chromatography with ultraviolet detection separates NAD+ from related nucleotides. Liquid chromatography-mass spectrometry offers higher specificity and can distinguish NAD+ from close analogs. Each method has trade-offs in sensitivity, throughput, and equipment needs, so reported values depend heavily on extraction and detection choices.
Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide built from adenine, nicotinamide, two ribose sugars, and two phosphate groups. The oxidized form carries a positive charge on the nicotinamide ring, while the reduced form, NADH, is neutral. This pair acts as a reversible electron carrier in cells. NAD+ is present in bacteria, plants, animals, and fungi. Its structure allows it to accept and donate electrons without being consumed in the reactions it supports.
In redox reactions, NAD+ accepts a hydride ion, which consists of two electrons and one proton. The hydride adds to the nicotinamide ring at a specific carbon, converting NAD+ into NADH. Dehydrogenase enzymes use this step in glycolysis, the citric acid cycle, and fatty acid oxidation. NADH later donates electrons to the mitochondrial electron transport chain, helping to drive ATP synthesis. The balance between NAD+ and NADH reflects the metabolic state of a cell, and shifts in that balance can alter how pathways operate.
The molecule was first described in the early twentieth century as a factor that promoted fermentation in yeast extracts. Later work linked it to hydrogen transfer and to the oxidation of nutrients in living tissues. Its structure was resolved as a dinucleotide, which explained why it could accept and donate electrons at specific enzyme sites. Today, NAD+ is recognized as a central substrate and signaling precursor, not merely a metabolic cofactor. Whether all observed NAD+ changes reflect causal signaling remains an open question.
Related compounds include NADH, the reduced form, and NADP+, which carries an additional phosphate group. NADP+ and NADPH often serve in biosynthetic and antioxidant reactions, while NAD+ and NADH are more associated with energy-yielding catabolism. Nicotinamide, nicotinic acid, and nicotinamide riboside are precursors that can enter salvage pathways. The exact contribution of dietary precursors to tissue NAD+ pools is an area of active investigation. Some studies measure labeled precursors to trace those routes.
NAD+ is the oxidized form of nicotinamide adenine dinucleotide, a coenzyme built from two nucleotides joined by a phosphate linkage. One nucleotide carries adenine, and the other carries nicotinamide; the plus sign denotes a formal positive charge on the nicotinamide ring, not a free proton. In cells, NAD+ and its reduced partner NADH form a reversible redox pair. That pair participates in electron transfer reactions throughout metabolism. The abbreviation NAD+ is common in biochemistry, while NAD(H) sometimes denotes the combined pool.
==== Type II antithrombin deficiency ==== Type II antithrombin deficiency is characterized by normal antithrombin levels but reduced antithrombin activity in the blood of affected individuals. It was originally proposed that type II deficiency be further divided into three subgroups (IIa, IIb, and IIc) depending on which antithrombin functional activity is reduced or retained.

== Evolution == The Capsicum genus split from Solanaceae 19.6 million years ago, 5.4 million years after the appearance of Solanaceae, and is native only to the Americas. Chilies only started to quickly evolve in the past 2 million years into markedly different species. This evolution can be partially attributed to a key compound found in peppers, 8-methyl-N-vanillyl-6-nonenamide, otherwise known as capsaicin. Capsaicin evolved similarly across species of chilies that produce capsaicin. Its evolution over the course of centuries is due to genetic drift and natural selection, across the genus Capsicum. Despite the fact that chilies within the Capsicum genus are found in diverse environments, the capsaicin found within them all exhibits similar properties that serve as defensive and adaptive features. Capsaicin evolved to preserve the fitness of peppers against fungi infections, insects, and granivorous mammals.
== Epidemiology == Catatonia has been historically studied in psychiatric patients. Catatonia is under-recognized because the features are often mistaken for other disorders, including delirium or the negative symptoms of schizophrenia. The prevalence has been reported to be as high as 10% in those with acute psychiatric illnesses, and 9–30% in the setting of inpatient psychiatric care. The incidence of catatonia is 10.6 episodes per 100 000 person-years, which essentially means that in a group of 100,000 people, the group as a whole would experience 10 to 11 episodes of catatonia per year. Catatonia can occur at any age, but is most commonly seen in adolescence or young adulthood or in older adults with existing medical conditions. It occurs in males and females in approximately equal numbers. Around 20% of all catatonia cases can be attributed to a general medical condition.
The 2014 State Department's annual Trafficking in Persons (TIP) report appeared to classify the Khobragade incident as an example of human trafficking, stating: "An Indian consular officer at the New York consulate was indicted in December 2013 for visa fraud related to her alleged exploitation of an Indian domestic worker." In response, India has shown no urgency to allow visits to India by the newly appointed US anti-human trafficking ambassador Susan P. Coppedge and the US special envoy for LGBT rights Randy Berry. Under Section 377 of the Indian Penal Code homosexuality was illegal in India. Indian Ambassador to the US, Arun K. Singh reiterated India's commitment to work within an international framework to tackle the problem of trafficking but rejected any "unilateral assessments" by another country saying "We will never accept it" and downplayed the importance of the visits: "When you ask a U.S. official when somebody will be given a visa, they always say 'we will assess when visa is applied for.' ... I can do no better than to reiterate the U.S. position." In February 2016, the Obama administration notified the US Congress that it intended to provide Pakistan eight nuclear-capable F-16 fighters and assorted military goods including eight AN/APG-68(V)9 airborne radars and eight ALQ-211(V)9 electronic warfare suites despite strong reservations from US lawmakers regarding the transfer of any nuclear weapons capable platforms to Pakistan. Shashi Tharoor, an elected representative from the Congress party in India, questioned the substance of India–U.S.

== Prognosis == AMC is considered non-progressive, so with proper medical treatment things can improve. The joint contractures will not get worse than they are at the time of birth. There is no way to resolve or cure AMC completely but with proper treatment most children make significant improvements in their range of motion and ability to move their limbs, which enables them to carry out daily activities and live relatively normal lives. Therapeutic interventions that are cornerstones in the treatment of AMC include: stretching and range of motion exercises, physical, occupational and speech therapy, splinting and serial casting. Surgical intervention may also improve joint mobility and function. Other positive prognostic factors for independent walking are active hips and knees, hip flexion contractures of less than 20 degrees and knee flexion contractures of less than 15 degrees without severe scoliosis.
These ultrafine particles are small enough to bypass respiratory defenses and be inhaled deep into the lungs, where they can enter the bloodstream and are associated with severe systemic inflammation, cardiovascular disease, and premature death. Many e-cigarette companies market their products as a smoking cessation aid without evidence of effectiveness. E-cigarette marketing has been found to make unsubstantiated health statements (e.g., that they help one quit smoking) including statements about improving psychiatric symptoms, which may be particularly appealing to smokers with mental illness. E-cigarette marketing advocate weight control and emphasize use of nicotine with many flavors. These marketing angles could particularly entice overweight people, young people, and vulnerable groups. Some e-cigarette companies state that their products are green without supporting evidence which may be purely to increase their sales.
==== Malaysia ==== Sup torpedo (lit. 'torpedo soup'; also sup hameed) is a Malaysian Indian cuisine bull penis curry soup, a delicacy of mamak stalls typically served with roti benggali. The soup is typically eaten as an aphrodisiac; variations include sup torpedo campur, which includes goat's testicles and horse penis. The Mufti of Federal Territory's Office opines that eating sup torpedo, although halal, is makruh for being genital meat.
==== Security ==== With the growing reliance on technology, there have been security and privacy concerns along with it. Billions of people use different online payment methods—such as WeChat Pay, PayPal, Alipay, and many more—to help transfer money. Although security measures are placed, some criminals are able to bypass them. In March 2022, North Korea used Blender.io, a mixer that helped them to hide their cryptocurrency exchanges, to launder over $20.5 million in cryptocurrency from Axie Infinity and steal over $600 million worth of cryptocurrency from the game's owner. Because of this, the U.S. Treasury Department sanctioned Blender.io, which marked the first time it has taken action against a mixer, to try to crack down on North Korean hackers. The privacy of cryptocurrency has been debated. Although many customers like the privacy of cryptocurrency, many also argue that it needs more transparency and stability.
A common and popular fish for tropical freshwater aquaria, C. macracanthus is a visually pleasing species. Although specimens in the wild will reach 40 to 50 cm, in the aquaria it is uncommon for C. macracanthus to attain more than 15 to 20 cm fork length. They are a very long-lived species, living up to 30 years. These fish have bifurcated spines under the eyes. They are thought to be used as a defence mechanism and possibly, for obtaining prey. C. macracanthus will associate with other benthic species in the aquaria if kept singly or with few other specimens. They make suitable tank-mates for any non-aggressive community fishes, but do not thrive when kept with larger, more dominant species. Occasionally C. macracanthus will exhibit erratic swimming patterns, such as swimming on their sides or upside down. This is not usually a sign of illness, however, and the fish will typically resume normal behaviour almost instantly. As C. macracanthus is found in riverine environments it is able to cope with good flow rates of water in the aquaria. Aquascapes may replicate this environment to suit the requirements of the species with areas of high flow, but also typically include areas of low flow to allow the fish to rest. C. macracanthus is also fond of cover and is not overly accustomed to bright lighting. The fish will often seek areas of shade or cover in which to rest during the day and become more active when light levels are more subdued. The barbels of clown loaches are known to be easily damaged by rough substrates, such as gravel, in aquaria.

Pancreatic beta cell function (synonyms Gβ or, if calculated from fasting concentrations of insulin and glucose, HOMA-Beta or SPINA-GBeta) is one of the preconditions of euglycaemia, i.e. normal blood sugar regulation. It is defined as insulin secretory capacity, i.e. the maximum amount of insulin to be produced by beta cells in a given unit of time.
=== Menkes disease === Menkes disease, a genetic condition of copper deficiency, was first described by John Menkes in 1962. It is a rare X-linked disorder that affects approximately 1/200,000 live births, primarily boys. Livers of Menkes disease patients cannot absorb essential copper needed for patients to survive. Death usually occurs in early childhood: most affected individuals die before the age of 10 years, although several patients have survived into their teens and early 20s. The protein produced by the Menkes gene is responsible for transporting copper across the gastrointestinal tract (GIT) mucosa and the blood–brain barrier. Mutational defects in the gene encoding the copper ATPase cause copper to remain trapped in the lining of the small intestine. Hence, copper cannot be pumped out of the intestinal cells and into the blood for transport to the liver and consequently to rest of the body. The disease therefore resembles a severe nutritional copper deficiency despite adequate ingestion of copper. Symptoms of the disease include coarse, brittle, depigmented hair and other neonatal problems, including the inability to control body temperature, intellectual disability, skeletal defects, and abnormal connective tissue growth. Menkes patients exhibit severe neurological abnormalities, apparently due to the lack of several copper-dependent enzymes required for brain development, including reduced cytochrome c oxidase activity. The brittle, kinky hypopigmented hair of steely appearance is due to a deficiency in an unidentified cuproenzyme.

== Mechanism of action == NGF binds with at least two classes of receptors: the tropomyosin receptor kinase A (TrkA) and low-affinity NGF receptor (LNGFR/p75NTR). Both are associated with neurodegenerative disorders. When NGF binds to the TrkA receptor, it drives the homodimerization of the receptor, which in turn causes the autophosphorylation of the tyrosine kinase segment. The tropomyosin receptor kinase A receptor has five extracellular domains, and the fifth domain is sufficient in binding NGF. Once bound, the complex undergoes endocytosis and activates the NGF transcriptional program, following two major pathways, the Ras/MAPK pathway and the PI3K/Akt pathway. The binding of NGF to TrkA also leads to the activation of PI 3-kinase, ras, and PLC signaling pathways. Alternatively, the p75NTR receptor can form a heterodimer with TrkA, which has higher affinity and specificity for NGF. Studies suggest that NGF circulates throughout the entire body via the blood plasma, and is important for the overall maintenance of homeostasis.

NAD+ is an oxidized dinucleotide coenzyme that carries electrons in metabolic reactions. It is also consumed by signaling enzymes, including sirtuins and PARPs. Its reduced form is NADH.
NAD+ is the oxidized form and can accept a hydride equivalent. NADH is the reduced form and donates electrons to the electron transport chain. The two forms cycle between each other during cellular respiration.
In mammals, NAD+ is synthesized mainly through salvage pathways using nicotinamide, nicotinamide riboside, or nicotinic acid. Tryptophan can also contribute through a de novo route. The salvage pathway is often considered the primary source in many tissues.
Aqueous NAD+ solutions are best kept frozen in aliquots and protected from light. Repeated freezing and thawing is avoided because it can accelerate breakdown. Dry powder stored desiccated at -20 °C or lower typically remains stable for longer periods.
