In humans, NAD+ can be synthesized from nicotinic acid, nicotinamide, nicotinamide riboside, and tryptophan through overlapping pathways. The salvage pathway recycles nicotinamide back to NAD+ and is often considered a major route in many tissues. Dietary precursors and intracellular recycling both contribute to the pool, but the quantitative importance of each source remains an active research question. NAD+ levels are not uniform across organs or cell compartments. Measurements in blood do not necessarily reflect concentrations inside tissues.
NAD+ is a dinucleotide composed of nicotinamide, ribose, and adenine linked by phosphate groups. Its full name is nicotinamide adenine dinucleotide, with "+" denoting the oxidized form. The molecule acts as a coenzyme in redox reactions, cycling between NAD+ and NADH. In cells, it participates in electron transfer during glycolysis, the citric acid cycle, and oxidative phosphorylation. It is distinct from NADP+, which carries an additional phosphate group and supports different biosynthetic reactions.
Beyond redox chemistry, NAD+ serves as a substrate for enzymes that transfer ADP-ribose or remove acetyl groups. Sirtuins, PARPs, and CD38-family enzymes consume NAD+ and produce nicotinamide and ADP-ribose-related products. These reactions link NAD+ availability to DNA repair, chromatin modification, and cellular signaling. Because the molecule is central to energy metabolism and regulation, changes in its concentration are studied in aging, immunity, and metabolic research. The balance between synthesis and consumption varies by tissue, developmental stage, and physiological state.
Commercial NAD+ is supplied as a solid, often as the free acid or a salt, and purity is verified by chromatographic methods. Laboratories typically store it desiccated at minus 20 degrees Celsius or below. Working solutions are prepared fresh because even sterile aqueous solutions can lose activity over hours to days depending on pH and temperature. Documentation may include a certificate of analysis, an assay value, and a recommended retest date. Researchers should verify identity and purity when results depend on precise cofactor concentrations.
NAD+ is commonly measured by high-performance liquid chromatography with ultraviolet detection, often at 254 or 260 nm. Enzymatic cycling assays provide higher sensitivity by coupling NAD+ to a reporter reaction. Mass spectrometry can distinguish NAD+ from close analogues and confirm isotope labeling. Sample preparation usually involves rapid quenching of metabolism to prevent interconversion with NADH. Because NAD+ and NADH differ by one hydride, extraction conditions strongly affect the measured ratio.
In aqueous solution, NAD+ is most stable under mildly acidic to neutral conditions and degrades faster at high pH or elevated temperature. The molecule can hydrolyze at the pyrophosphate bond or undergo nonenzymatic cyclization. Buffers, chelating agents, and cold temperatures slow these losses during analysis. Repeated freeze-thaw cycles are generally avoided because they can promote degradation and concentration changes. Light exposure is also controlled, though NAD+ is less photolabile than some related nucleotides.

Commercial NAD+ is available at research grade, often with purity specifications determined by high-performance liquid chromatography. Certificates of analysis may report water content, residual solvents, and counterion identity. Identity can be confirmed by ultraviolet absorbance near 260 nm, mass spectrometry, or enzymatic activity. Because different salt forms and hydration states exist, researchers should verify that the product matches the intended molecular form. Lot-to-lot variation in purity can affect quantitative assays and should be documented.
Solid NAD+ is usually supplied as a white to off-white powder or lyophilized preparation. It is hygroscopic and should be kept desiccated at low temperature, commonly -20 °C or below for long-term storage. Aqueous solutions are less stable than dry material and are often prepared fresh or stored frozen in aliquots. Light exposure and repeated freeze-thaw cycles can promote degradation, so amber containers and single-use aliquots are preferred. Buffered solutions near neutral pH are generally less stable than acidic or frozen preparations.
Quantification of NAD+ in biological samples typically uses liquid chromatography coupled to mass spectrometry. Enzymatic cycling assays offer higher throughput and rely on NAD+ dependent dehydrogenases to amplify signal. Both approaches require careful sample quenching because NAD+ can be rapidly consumed or converted after collection. Acidic extraction is common for NAD+, while alkaline conditions favor NADH in some protocols. Isotopically labeled internal standards help correct for losses during extraction and ionization.
The molecule was first described in the early twentieth century as a factor that promoted fermentation in yeast extracts. Later work linked it to hydrogen transfer and to the oxidation of nutrients in living tissues. Its structure was resolved as a dinucleotide, which explained why it could accept and donate electrons at specific enzyme sites. Today, NAD+ is recognized as a central substrate and signaling precursor, not merely a metabolic cofactor. Whether all observed NAD+ changes reflect causal signaling remains an open question.
Related compounds include NADH, the reduced form, and NADP+, which carries an additional phosphate group. NADP+ and NADPH often serve in biosynthetic and antioxidant reactions, while NAD+ and NADH are more associated with energy-yielding catabolism. Nicotinamide, nicotinic acid, and nicotinamide riboside are precursors that can enter salvage pathways. The exact contribution of dietary precursors to tissue NAD+ pools is an area of active investigation. Some studies measure labeled precursors to trace those routes.
NAD+ is the oxidized form of nicotinamide adenine dinucleotide, a coenzyme built from two nucleotides joined by a phosphate linkage. One nucleotide carries adenine, and the other carries nicotinamide; the plus sign denotes a formal positive charge on the nicotinamide ring, not a free proton. In cells, NAD+ and its reduced partner NADH form a reversible redox pair. That pair participates in electron transfer reactions throughout metabolism. The abbreviation NAD+ is common in biochemistry, while NAD(H) sometimes denotes the combined pool.
Measuring NAD+ in biological samples requires rapid processing because the compound can degrade or interconvert after collection. Common approaches include enzymatic cycling assays, high-performance liquid chromatography, and mass spectrometry. Each method has different sensitivity, specificity, and susceptibility to interference from related nucleotides. Sample type matters: cultured cells, animal tissues, and human blood present distinct challenges. Reported values can vary widely across laboratories because of differences in extraction, normalization, and analytical platform. Standardization remains an open issue in the field.
NAD+ is relatively unstable in aqueous solution, especially at neutral or alkaline pH and at elevated temperatures. It is typically stored dry, protected from light and moisture, and kept cold or frozen for long-term use. Solutions are often prepared fresh or buffered to mildly acidic pH to slow hydrolysis. Repeated freeze-thaw cycles can reduce integrity. Laboratories may verify concentration using ultraviolet absorbance at 259 nm or by enzymatic assay. These handling practices are general laboratory conventions rather than universal rules.
Beyond redox chemistry, NAD+ acts as a substrate for several enzyme families. ADP-ribosyltransferases, sirtuins, and CD38 ectoenzymes cleave the molecule into nicotinamide and ADP-ribose or related products. These reactions connect NAD+ availability to processes such as DNA repair, chromatin modification, and calcium signaling. Because the coenzyme is used in both electron transfer and signaling, cells maintain separate pools in compartments including the cytosol, mitochondria, and nucleus. The relative sizes and regulation of those pools remain active areas of study.
Cells produce NAD+ through several biosynthetic routes. The salvage pathway recycles nicotinamide, while the Preiss-Handler pathway uses nicotinic acid, and a de novo route can start from tryptophan in some organisms. In mammals, the salvage pathway is generally considered the main source under ordinary conditions. Tissue concentrations vary widely by cell type and compartment, and measured declines with age have been reported in some studies. Whether such changes drive aging or mainly accompany it remains an open question.
Nicotinamide adenine dinucleotide, commonly abbreviated NAD+, is a coenzyme present in all living cells. The molecule consists of two nucleotides linked by phosphate groups, with adenine and a nicotinamide ring as its principal features. In its oxidized form, the nicotinamide ring can accept a hydride ion, becoming NADH. This reversible conversion places NAD+ at the center of many electron-transfer reactions. Its role as a redox carrier is well established across bacteria, plants, fungi, and animals.
==== Utility ==== The main clinical utility of NT-proBNP is that a normal level helps to rule out chronic heart failure in the emergency setting. An elevated NT-proBNP should never be used exclusively to "rule in" acute or chronic heart failure in the emergency setting due to lack of specificity . Either BNP or NT-proBNP can also be used for screening and prognosis of heart failure. Increased NT-proBNP adjusted for age and sex and annual increase of NT-proBNP above 50% are associated with increased event rate in patients with non-severe aortic valve stenosis. BNP and NT-proBNP are also typically increased in patients with left ventricular dysfunction, with or without symptoms (BNP accurately reflects current ventricular status, as its half-life is 20 minutes, as opposed to 1–2 hours for NT-proBNP). A preoperative BNP can be predictive of a risk of an acute cardiac event during vascular surgery. A cutoff of 100 pg/ml has a sensitivity of approximately 100%, a negative predictive value of approximately 100%, a specificity of 90%, and a positive predictive value of 78% according to data from the United Kingdom. There is a diagnostic 'gray area', often defined as between 100 and 500 pg/mL, for which the test is considered inconclusive, but, in general, levels above 500 pg/ml are considered to be an indicator of heart failure. This so-called gray zone has been addressed in several studies, and using clinical history or other available simple tools can help make the diagnosis. Some laboratories report in units ng per Litre (ng/L), which is equivalent to pg/mL.
=== Australia === In Australia, the supply of kava is regulated through the National Code of Kava Management. Travellers to Australia are allowed to bring up to 4 kg of kava in their baggage, provided they are at least 18 years old and the kava is in root or dried form. Commercial import of larger quantities is allowed under licence for medical or scientific purposes. These restrictions were introduced in 2007 after concern was raised about abuse of kava in indigenous communities. Initially, the import limit was 2 kg per person; it was raised to 4 kg in December 2019, and a pilot program allowing for commercial importation was implemented on 1 December 2021. The Australian Therapeutic Goods Administration has recommended no more than 250 mg of kavalactones be taken in a 24‑hour period. Kava possession is limited to 2 kg per adult in the Northern Territory. While it was previously banned in Western Australia, the Western Australian Health Department lifted the ban in February 2017, bringing Western Australia "into line with other States" where it has always remained legal, albeit closely regulated.
== Contraindications == The drug is contraindicated in people with known hypersensitivity to ertapenem or other carbapenem type antibiotics, or with severe hypersensitivity reactions (such as anaphylaxis or severe skin reactions) to other beta-lactam antibiotics in the past.
In addition, a very thorny issue was that of "replacements", a compensation that Peru would make to Colombian troops for the casualties suffered by them during the war, by exchanging Peruvian citizens and expatriating them to Gran Colombia, exorbitant and inhuman requirement, given that the war had already ended. Given this, Bolívar demobilized most of the Peruvian units (about five thousand who replaced the Gran Colombian casualties) and after the capitulation of Callao, presented on February 10, 1826, the demand to send 6,000 Peruvian recruits to serve Venezuela, sending the first contingents in July, probably no more than 3,000, officially to reinforce the defenses against a possible French invasion (Cien Mil Hijos de San Luis) but in reality it was to confront General Páez, who had started La Cosiata (a separatist movement in Venezuela) and also to demilitarize Peru so that it is not a future threat to its continental projects in the Patria Grande. The troops sent there, due to the distance and lack of knowledge of the language (the majority were indigenous who barely knew Spanish) made desertion or mutiny difficult, many of those sent dying due to the weather and tropical diseases. It is known that some survivors were repatriated of New Granada and Venezuela in 1852 and 1857 respectively. Due to the lifelong and authoritarian constitutions promulgated for Peru and Bolivia, the Liberator would be accused of using his armies to impose political systems, in a republican imperialism, rather than making countries independent.
==== Silver ==== A tarnished-silver or aluminium paint-like feces color characteristically results when biliary obstruction of any type (white stool) combines with gastrointestinal bleeding from any source (black stool). It can also suggest a carcinoma of the ampulla of Vater, which will result in gastrointestinal bleeding and biliary obstruction, resulting in silver stool.

=== Return to Biotin receptor isolation === Hofmann came full circle to his early work on biotin when he attached this vitamin to insulin. He spent a sabbatical leave in Aachen in Helmut Zahn's laboratory to learn techniques for modifying insulin). Using this information, he was able to chemically attach biotin to one of the three Lys residues of the insulin chains, thus producing an insulin that would bind to columns of avidin-Sepharose. The biotinyl-insulin receptor complex could then be displaced by biotin. With this tool, Hofmann and his coworkers successfully isolated a fully active insulin receptor. His final work was directed toward isolation of the ACTH receptor using the same approach used on the insulin receptor but by this time his health was in decline.
== Disease relevance == MMP-3 has been implicated in exacerbating the effects of traumatic brain injury (TBI) through its disruption of the blood-brain barrier (BBB). Different studies have shown that after the brain undergoes trauma and inflammation has begun, MMP production in the brain is increased. In a study conducted using MMP-3 wild type (WT) and knockout (KO) mice, MMP-3 was shown to increase BBB permeability after traumatic injury. The WT mice were shown to have lower claudin-5 and occludin levels than the KO mice after TBI. Claudin and occludin are proteins that are essential for the formation of the tight junctions between the cells of the blood-brain barrier. Tissue from uninjured WT and KO mice brains was also treated with active MMP-3. Both the WT and KO tissues showed a drop in claudin-5, occludin, and laminin-α1 (a basal lamina protein), suggesting that MMP-3 directly destroys tight junction and basal lamina proteins. MMP-3 also does damage to the blood-spinal cord barrier (BSCB), the functional equivalent of the blood-brain barrier, after spinal cord injury (SCI). In a similar study conducted using MMP-3 WT and KO mice, MMP-3 was shown to increase BSCB permeability, with the WT mice showing greater BSCB permeability than the KO mice after spinal cord injury. The same study also found decreased BSCB permeability when spinal cord tissues were treated with a MMP-3 inhibitor. These results suggest that the presence of MMP-3 serves to increase BSCB permeability after SCI.
Integrin alpha-2, or CD49b (cluster of differentiation 49b), is a transmembrane protein which in humans is encoded by the CD49b gene. The CD49b protein is an integrin alpha subunit. It makes up half of the α2β1 integrin duplex. Integrins are heterodimeric integral membrane glycoproteins composed of a distinct alpha chain and a common beta chain. They are found on a wide variety of cell types including T cells (the NKT cells), NK cells, fibroblasts and platelets. Integrins are involved in cell adhesion and also participate in cell-surface-mediated signalling. The α2β1 integrin functions primarily as a recepter for collagen, making it important in tissues like the skin, bone, and blood vessels. Expression of CD49b in conjunction with LAG-3 has been used to identify type 1 regulatory (Tr1) cells. The DX5 monoclonal antibody recognizes mouse CD49b.
All known isotopes of curium are radioactive and have small critical mass for a nuclear chain reaction. The most stable isotope, 247Cm, has a half-life of 15.6 million years; the longest-lived curium isotopes predominantly emit alpha particles. Radioisotope thermoelectric generators can use the heat from this process, but this is hindered by the rarity and high cost of curium. Curium is used in making heavier actinides and the 238Pu radionuclide for power sources in artificial cardiac pacemakers and RTGs for spacecraft. It served as the α-source in the alpha particle X-ray spectrometers of several space probes, including the Sojourner, Spirit, Opportunity, and Curiosity Mars rovers and the Philae lander on comet 67P/Churyumov–Gerasimenko, to analyze the composition and structure of the surface.

==== Initial campaigns (October–December 1932) ==== Liu Wenhui was taken off-guard by the sudden war; in contrast, Liu Xiang had developed a strategy of cutting his uncle's supply lines. Liu Xiang's forces would first attack Luzhou in a combined arms offensive featuring his army, navy, and aviation, capturing the city. After the loss of Luzhou, an uprising broke out in Yibin against the rule of Liu Wenhui and his extractive tax collector and brother Liu Wencai. Liu Wencai took advantage of the chaos gripping the city to loot around 1.4 million silver dollars, even taking artifacts from the Luzhou Fuzhou Hall. Liu Wencai then escaped, disguised as a foreigner, ending his rule of Yibin. Thus, already caught off-balance, Liu Wenhui lost two of his most important cities at the very start of the war. Liu Xiang's army then began advancing towards Leshan. In November, Liu Wenhui would end up antagonizing the other Baoding warlords by attacking Tian Songyao's forces in Chengdu. The ensuing urban warfare caused over 20,000 casualties and 27,000 refugees, leading to both warlords losing much of their popular support. Deng Xihou, the third Baoding warlord, attempted to mediate the conflict, but with no success. Although Tian Songyao had been defeated by December and even offered to retire, his subordinates refused to let him step down. In the end, a ceasefire was reached and Tian remained in Chengdu, causing no gain for Liu Wenhui, who had departed for Meishan to set up his headquarters.
==== Other optical imaging ==== For in vivo fluorescence-based optical imaging, dyes that emit near infrared (NIR) wavelengths >700 nm are most effective, such as indocyanine green, but encounter limitations with reduced circulation time and nonspecific interactions with other biological factors that affect the fluorescence. pH-sensitive nanogels with functionalized surface receptors to target cancer cells were loaded with a fluorescent dye that was only released upon endocytosis. These nanogels successfully generated a fluorescent signal from within the cancer cells and many other groups have developed similar technologies.
These social and economic realities do not, however, diminish the fact that the Cold War was a "multi-dimensional struggle" that left physical remnants across Europe, from "missile silos, tank tracks, command bunkers, and troop barracks" to the stockpiling of nuclear armaments capable of destroying the entire planet. Jarausch adds that:
Burroughs found Trump's efforts to freeze billions of dollars of funding for Harvard illegal, writing that the government had infringed upon Harvard's free speech rights and that it was "difficult to conclude anything other than that defendants used antisemitism as a smokescreen for a targeted, ideologically-motivated assault on this country's premier universities".

The plus sign indicates the oxidized form of nicotinamide adenine dinucleotide, which can accept electrons. When it accepts electrons, it becomes NADH. The two forms together support redox reactions in cells.
No. NAD+ is the oxidized form and NADH is the reduced form. They differ by two electrons and a proton equivalent, and cells interconvert them during metabolism.
Yes. NAD+ is present in all living cells and is required for fundamental metabolic reactions. Its concentration varies by tissue, compartment, and time.
NAD+ and NADH can interconvert quickly after a sample is collected, which can alter the measured ratio. Rapid quenching and cold handling limit enzymatic and chemical changes.
